Target Information

Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are transported within the cell by cation-independent mannose 6-phosphate receptors (CI-MPR, ~300 kDa).
MPRs function in two interconnected cycles:

  1. Biosynthetic cycle: Transports newly synthesized lysosomal enzymes from the trans Golgi network to late endosomes or early lysosomes.
  2. Endocytic cycle: Transports extracellular lysosomal enzymes from the plasma membrane via clathrin-coated vesicles to early endosomes.

MPRs cycle between these compartments approximately every 3 hours. Most are located in late endosomes, fewer in the trans Golgi network, and about 10% at the cell surface.
In addition to man6P binding, MPRs possess a separate binding site for type II insulin-like growth factor (IGF-II) and can bind both ligands simultaneously.
A soluble ~240 kDa truncated form, representing the extracellular domain, circulates in serum and retains ligand-binding capability.
