Target Information

The second messengers cAMP and cGMP are key regulatory molecules involved in various signal transduction pathways. Their levels are controlled by synthesis via nucleotide cyclases and hydrolysis by cyclic nucleotide phosphodiesterases (PDEs). PDEs form a superfamily of enzymes that convert 3',5'-cyclic nucleotides to nucleoside 5'-monophosphates.
PDE6 functions as the effector enzyme in the G protein-mediated signal transduction cascade in the visual system. It consists of several subunits: rod and cone-specific catalytic subunits (alpha', alpha, beta), the inhibitory subunit gamma, and subunit delta of unknown function. The catalytic core is formed by alpha'/alpha' homodimers in cones and alpha/beta heterodimers in rods. The C-terminus of both catalytic and inhibitory subunits undergoes methylation, myristoylation, and prenylation, which are essential for complex assembly and membrane association.
