Target Information
NLK is a serine/threonine kinase involved in signal transduction and cellular communication. It acts as a positive effector of the non-canonical Wnt signaling pathway, functioning downstream of WNT5A, MAP3K7/TAK1, and HIPK2. Activation leads to phosphorylation of the histone methyltransferase SETDB1, forming a complex that interacts with PPARG, resulting in transcriptional silencing of PPARG target genes. This process inhibits adipogenesis and promotes osteoblastogenesis in mesenchymal stem cells.
NLK also negatively regulates the canonical Wnt/β-catenin and Notch signaling pathways by phosphorylating key transcription factors, thereby modulating gene expression.
