Target Information
MLK4 (Mixed Lineage Kinase 4) belongs to the MAP kinase kinase kinase (MAP3K) superfamily, containing both serine/threonine and tyrosine kinase activities in its catalytic domains. The kinase structure includes an N-terminal Src homology (SH3) domain, a kinase domain, a leucine zipper region, and a CDC42/RAC-interactive binding (CRIB) motif, with divergent C-terminal regions. MLK4 is highly expressed in kidney and pancreas and functions as a negative regulator of TLR4. It does not activate JNK1/MAPK8, p38/MAPK14, or ERK2/MAPK1 pathways.
