Product Specific Information
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. CD3e, along with gamma and delta subunits, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking via immobilized UCHT1 triggers intracellular signaling leading to activation and proliferation.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells. Recommended use: 5 µL (0.4 µg)/test (100 µL final volume).
Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Use 5 µL per sample for optimal staining with NovaFluor conjugates. Recommended to add to antibody cocktails before mixing with cells.
NovaFluor Technology:
Based on Phiton technology utilizing nucleic acid dye structures engineered to minimize spillover and spread impacts.
Excitation: 552 nm; Emission: 612 nm; Laser: 561 nm (Yellow).
Target Information
The CD3 complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits encoded on chromosome 11. CD3 is expressed on most T lymphocytes and thymocytes but not on B or NK cells. It plays roles in TCR-induced activation, proliferation, and signal transduction. Defects in CD3 genes are linked to immunodeficiency.
