The Human Granzyme A Simplex ProcartaPlex Kit measures granzyme A protein and is designed to be combinable with other Simplex kits, allowing creation of custom multiplex panels using Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits, only one buffer kit (sold separately) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on sandwich ELISA principles, using two highly specific antibodies binding to different epitopes of a single protein to quantitate multiple targets simultaneously via a Luminex instrument. These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and yield results in approximately four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample from plasma, serum, cell culture supernatants, and other bodily fluids.

Each Luminex bead in the ProcartaPlex assay is internally dyed with red and infrared fluorophores to create unique spectral signatures identifiable by Luminex xMAP systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX). Similar to a sandwich ELISA, matched antibody pairs identify the target protein. In multiplexed assays, each bead is labeled with antibodies specific for a single target protein, and bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).

Unlike traditional ELISA, ProcartaPlex assays use bead-conjugated capture antibodies, allowing reagents to remain free-floating in solution. The Luminex instrument employs two lasers: one to distinguish bead signatures and another to quantify RPE fluorescence, which correlates with protein concentration. This enables profiling of more target proteins using significantly less sample in comparable time to ELISA.
