The Human IL-12/IL-23p40 Simplex ProcartaPlex Kit measures IL-12/IL-23p40 protein and is designed to be combinable with other Simplex kits to create a customized multiplex panel utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits, only one buffer kit (sold separately) is required per assay plate, regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the principles of sandwich ELISA, using two highly specific antibodies binding to different epitopes of a protein to quantitate multiple targets simultaneously using a Luminex instrument. The assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, with results obtainable in approximately four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample. Luminex beads are internally dyed with precise proportions of red and infrared fluorophores to create unique spectral signatures identifiable by Luminex instruments (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).

Similar to a sandwich ELISA, matched antibody pairs identify the protein of interest. Each bead is labeled with antibodies specific for a single target protein, and bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The conjugation of antibodies to distinct beads allows multiplex analysis in a single well.

Unlike traditional ELISA, the capture antibody in ProcartaPlex assays is conjugated to a bead rather than adsorbed to a microplate well, allowing reagents to remain free-floating. Detection involves two lasers in the Luminex instrument—one to identify bead signatures and another to quantify RPE fluorescence proportional to protein concentration. This enables profiling of more proteins using less sample in comparable time to ELISA.
