Product Specific Information

This 3DS223H monoclonal antibody recognizes human CD223 (LAG-3), a 70-kDa surface glycoprotein belonging to the Ig superfamily with homology to CD4.
LAG-3 binds to MHC class II with higher affinity than CD4 and negatively regulates T cell activation and homeostatic proliferation.
It is expressed on activated T cells (including regulatory T cells) and NK cells, with CD8+ T cells showing higher levels than CD4+ T cells.
Coexpression of LAG-3 and CD49b identifies Type 1 regulatory T cells (Tr1 cells).

This antibody recognizes a formaldehyde-fixed epitope.
Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on stimulated normal human peripheral blood cells.
Recommended usage: 5 µL (0.06 µg) per test (100 µL total volume, 10⁵–10⁸ cells/test).

Note: NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining and should be added to antibody cocktails prior to combining with cells.
Use 5 µL per sample for both multi-color panels and single-color controls.

NovaFluor conjugates utilize Phiton™ technology, engineered for optimized fluorescence signatures minimizing spillover and spread.
Excitation: 639 nm; Emission: 700 nm; Laser: 633–640 nm (Red).
