Target Information
Sclerostin은 주로 골세포(osteocytes)에서 분비되는 당단백질로, Cerberus/DAN 계열의 BMP 억제 단백질에 속하며 Wnt 신호전달 경로의 내인성 조절자로 작용합니다.
이 단백질은 뼈, 골수, 연골, 신장, 간 등에서도 발현되며, 과형성 연골세포 및 cementocyte에서도 생성됩니다.
Sclerostin은 DKK-1, DKK-4와 유사하게 LRP5 및 LRP6과 복합체를 형성하여 Wnt/β-catenin 신호를 억제합니다. 또한 LRP4와 결합하여 Wnt 신호 억제를 매개하고, RANKL 발현을 증가시켜 파골세포 활성을 촉진할 수 있습니다.
Sclerostin의 뼈 형성과 흡수 조절 기능은 sclerosteosis 및 van Buchem disease(VBD)와 같은 희귀 골형성 이상 질환에서 그 중요성이 강조됩니다. 이러한 질환은 sclerostin 결핍으로 인한 과도한 골형성과 과광물화로 특징지어집니다.
