Target Information

Vacuolar-type H⁺-ATPase (V-ATPase) is a multisubunit enzyme responsible for acidification of eukaryotic intracellular organelles. V-ATPases pump protons against an electrochemical gradient, while F-ATPases reverse the process to synthesize ATP.
The enzyme consists of:

  • V1 domain: Responsible for ATP hydrolysis (subunits A–H)
  • V0 domain: Responsible for proton translocation (subunits a, d, c, c′, c″)

V-ATPase likely operates through a rotary mechanism.
The V1 B subunit exists as two isoforms:

  • B1 isoform: Functions in proton secretion in the inner ear, maintaining endolymph pH and auditory function.
  • B2 isoform: Expressed in kidney and osteoclasts.
    Mutations in the human B1 isoform gene (chromosome 2cen–q13) cause distal renal tubular acidosis associated with sensorineural deafness.
