Target Information
Peroxiredoxin (Prx) is a growing peroxidase family, whose mammalian members are known to be associated with cell proliferation, differentiation, and apoptosis. Many isoforms (about 50 proteins), grouped by amino acid sequence homology, contain active site cysteine residues and exhibit thiol-specific antioxidant activity, distributed throughout all kingdoms.
Mammalian Prx consists of six different members classified into typical 2-Cys, atypical 2-Cys, and 1-Cys Prx. Except for Prx VI (1-Cys subgroup), the other five 2-Cys Prx isotypes exhibit thioredoxin-dependent peroxidase (TPx) activity using thioredoxin, thioredoxin reductase, and NADPH as a reducing system.
Mammalian Prxs are 20–30 kDa in molecular size and vary in subcellular localization:
- Prx I, II, VI: Cytosol
- Prx III: Mitochondria
- Prx IV: Endoplasmic reticulum and secretion
- Prx V: Peroxisome, mitochondria, and cytosol
