Target Information
The Ragulator complex is composed of three proteins—C11orf59, ROBLD3, and MAPKSP1—which target mTORC1 to lysosomal surfaces. This endosomal trafficking mechanism is exploited by certain viruses, including coronavirus, which shows increased replication when C11orf59 is disrupted.
C11orf59 is positively regulated by cholesterol depletion and negatively regulated by cholesterol loading, playing a role in LDL uptake and cholesterol homeostasis. Together with PPAR-alpha, it influences lipid and carbohydrate metabolism in diabetes. Most effects of C11orf59 are associated with mTORC1 kinase–mediated growth promotion, and dysregulation of this pathway can affect normal cellular metabolism.
