Recombinant Human IFN-gamma produced in E. coli is a single, non-glycosylated polypeptide chain containing 144 amino acids with a molecular mass of approximately 16,879 Da.
IFN-gamma, also known as Type II interferon, is a homodimeric glycoprotein with subunits of approximately 21–24 kDa. The human IFN-gamma gene, located on chromosome 12, contains three introns and four exons encoding a 166-amino-acid polypeptide, including a 20-amino-acid signal peptide.
Unlike IFN-alpha and IFN-beta, which can be produced by various cell types, IFN-gamma is secreted primarily by T cells and NK cells. It exhibits antiviral and antiparasitic activities, inhibits proliferation of normal and transformed cells, and acts synergistically with TNF-alpha and TNF-beta in growth inhibition.

Target Information
IFN-gamma (Interferon gamma, Type II interferon) is a macrophage activation factor and immune interferon produced mainly by T-lymphocytes and natural killer (NK) cells in response to antigens, mitogens, and cytokines.
It is a dimeric glycoprotein consisting of two 146-amino-acid subunits, forming a functional homodimer of approximately 45 kDa. On SDS-PAGE, IFN-gamma appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.
The biological activity of IFN-gamma is highly species-specific; human IFN-gamma does not cross-react with mouse IFN-gamma.
Functions include:
- Antiviral and tumor antiproliferative activity
- Induction of MHC class I and II expression
- Macrophage activation
- Enhancement of immunoglobulin secretion by B lymphocytes
- Cytokine regulation and synergistic interaction with other cytokines
Activation occurs through binding to IFN-gamma receptor I and II, initiating the JAK-STAT signaling pathway. Human IFN-gamma shares approximately 40% sequence homology with mouse IFN-gamma and is upregulated by IL-2, FGF-basic, and EGF, while downregulated by vitamin D3 or DMN. Mutations in the IFN-gamma gene have been associated with aplastic anemia.
