Low density lipoprotein-related protein 2 (LRP2), also known as megalin, is a multi-ligand endocytic receptor expressed in various tissues, primarily in absorptive epithelial tissues such as the kidney. This glycoprotein contains a large extracellular domain, a single transmembrane domain, and a short cytoplasmic tail. It binds diverse macromolecules including albumin, apolipoproteins B and E, and lipoprotein lipase. LRP2 is essential for the reuptake of ligands such as lipoproteins, sterols, vitamin-binding proteins, and hormones. It also participates in cell signaling, interacting with ligands such as parathyroid hormones and sonic hedgehog extracellularly, and MAP kinase scaffold proteins and JNK interacting proteins intracellularly. Phosphorylation of its cytoplasmic domain regulates receptor recycling. Mutations in this gene are associated with Donnai-Barrow syndrome (DBS) and facio-oculoacoustico-renal syndrome (FOAR).
