B4GALT1 (Beta-1,4-Galactosyltransferase 1) is a protein-coding gene involved in transport to the Golgi and subsequent modification and metabolism.
B4galt1 is one of seven beta-1,4-galactosyltransferase (beta4GalT) genes encoding type II membrane-bound glycoproteins with specificity for UDP-galactose.
The Golgi complex form catalyzes lactose production in lactating mammary glands and contributes to the synthesis of complex-type N-linked oligosaccharides in glycoproteins and glycolipids.
The cell surface form functions as a recognition molecule during cell-to-cell and cell-to-matrix interactions, such as development and fertilization, by binding specific oligosaccharide ligands.


Target Information

This gene is one of seven beta-1,4-galactosyltransferase (beta4GalT) genes encoding type II membrane-bound glycoproteins specific for UDP-galactose.
All transfer galactose in a beta1,4 linkage to acceptor sugars such as GlcNAc, Glc, and Xyl.
Each beta4GalT has distinct functions in glycoconjugate and saccharide biosynthesis.
These type II membrane proteins possess an N-terminal hydrophobic signal sequence that directs them to the Golgi apparatus and remains as a transmembrane anchor.

By sequence similarity, the beta4GalTs form four groups:

  • beta4GalT1 and beta4GalT2
  • beta4GalT3 and beta4GalT4
  • beta4GalT5 and beta4GalT6
  • beta4GalT7

This gene is unique among beta4GalT genes, encoding an enzyme participating in both glycoconjugate and lactose biosynthesis.
In glycoconjugate synthesis, the enzyme adds galactose to N-acetylglucosamine residues on glycoprotein chains.
In lactating mammary tissues, it forms a heterodimer with alpha-lactalbumin to catalyze UDP-galactose + D-glucose ⇔ UDP + lactose.
These two enzymatic forms arise from alternate transcription initiation and post-translational processing.
