Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, mature T helper cells, and monocytes. CD4 acts as a receptor for HIV; SK3 blocks HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 recognize distinct epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on human peripheral blood cells, used at 5 µL (0.1 µg)/test (final volume 100 µL, 10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread.

Target Information

CD4 is involved in recognition of MHC class II molecules and functions as a co-receptor for HIV. It is mainly expressed in T helper cells, monocytes, macrophages, and dendritic cells. Tissue-level expression includes thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. CD4 mediates early T-cell activation via association with TCR complex and Lck kinase, and may contribute to neuronal damage in infectious or immune-mediated CNS diseases.
