Target Information
PTEN은 다양한 암에서 높은 빈도로 변이되는 종양 억제 유전자로 알려져 있습니다. PTEN 유전자는 phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase를 암호화하며, tensin-like domain과 dual specificity protein tyrosine phosphatases와 유사한 촉매 도메인을 포함합니다.
이 단백질은 phosphoinositide 기질을 탈인산화하여 세포 내 phosphatidylinositol-3,4,5-trisphosphate 수준을 음성 조절하고, AKT/PKB 신호전달 경로를 억제함으로써 종양 억제 기능을 수행합니다.
또한 PTEN은 신경발생 과정에서 신경세포의 위치, 수상돌기 발달, 시냅스 형성 등을 조절하는 AKT-mTOR 경로의 핵심 조절자 역할을 합니다.
지방 조직에서 인슐린 신호 및 포도당 대사를 음성 조절할 수 있으며, 핵 내 단일 유비퀴틴화 형태는 더 높은 세포자멸 유도 능력을 나타냅니다.
