Target Information
The HPV E7 proteins are small zinc-binding phosphoproteins (HPV16 E7: 98 amino acids, 15 kDa) localized in the nucleus. They are structurally and functionally similar to the E1A protein of subgenus C adenoviruses. The N-terminal 16 amino acids of HPV16 E7 share homology with conserved region 1 (CR1) of E1A, and the next domain (up to amino acid 37) is homologous to region 2 (CR2) of E1A. These domains are essential for cell transformation in vitro, indicating mechanistic similarities between these viruses. The CR2 region contains the LXCXE motif (residues 22–26) that binds to the tumor suppressor protein pRb.
