Target Information
FAM175B is a component of the BRISC complex, a multiprotein complex that specifically cleaves Lys-63-linked polyubiquitin, retaining the last ubiquitin on substrates. It acts as a scaffold protein maintaining BRISC components in the cytoplasm and regulates apoptosis by modulating Lys-63-linked ubiquitination. FAM175B is essential for mitotic spindle assembly and kinetochore attachment via deubiquitination of NUMA1. It also regulates interferon signaling through IFNAR1 deubiquitination, stabilizing IFNAR1 and enhancing its activity. Additionally, it down-regulates LPS responses and is required for proper p53 induction following DNA damage, promoting USP7–p53 interaction and preventing p53 degradation.
