Product Specific Information
Immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, representing about 10% of lymphocyte surface proteins.
It is absent on non-hematopoietic cell lines and tissues. CD45 regulates Src-family kinases and plays a key role in lymphocyte development and antigen signaling.
Multiple isoforms arise from alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
CD45RA is one isoform with restricted expression among lymphoid subtypes.
CD45 influences T- and B-cell receptor signaling, cytokine receptor regulation via JAK kinases, and integrin-mediated survival pathways.
