Product Specific Information
The IM7 monoclonal antibody reacts with all isoforms of mouse CD44 (Pgp-1). CD44 is expressed by hematopoietic and non-hematopoietic cells, with high expression in bone marrow myeloid cells and memory T cells. Peripheral B and T cells can upregulate CD44 expression. CD44 functions as an adhesion molecule via binding to hyaluronate, an extracellular matrix component.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications
- Reported for use in flow cytometric analysis
- Tested on mouse bone marrow cells and splenocytes
- Recommended use: ≤ 0.6 µg/test (100 µL final volume)
- Cell number: 10⁵–10⁸ cells/test (empirically determined)
- Titrate for optimal performance
Important Notes
- NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
- Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
- Add 5 µL per sample (regardless of panel size).
- For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information
CD44 is a 100 kDa type I transmembrane glycoprotein expressed on human leukocytes, white matter of the brain, and some epithelial cells (intestine, breast).
It serves as a receptor for hyaluronic acid (HA) and plays roles in:
- Cell-cell interactions
- Cell adhesion and migration
- Lymphocyte activation, recirculation, and homing
CD44 expression may increase in certain carcinomas, potentially linked to metastatic potential.
Multiple isoforms exist due to alternative splicing, contributing to its structural and functional diversity.
Diseases associated with CD44 dysfunction include superficial keratitis and lichen sclerosus.
