Target Information
ABRO (FAM175B) is a component of the BRISC complex, which specifically cleaves Lys-63-linked polyubiquitin chains while retaining the last ubiquitin on substrates. It acts as a scaffold for BRISC complex assembly and cytoplasmic retention. ABRO is involved in:
- Regulation of apoptosis via Lys-63-linked ubiquitination.
- Mitotic spindle assembly and kinetochore attachment through deubiquitination of NUMA1.
- Interferon signaling via deubiquitination of IFNAR1, enhancing receptor stability and surface expression.
- Down-regulation of LPS response through IFNAR1 modulation.
- Induction of p53/TP53 in response to DNA damage by promoting USP7–p53 interaction, leading to p53 stabilization and apoptosis.
