The Human CD62E (E-selectin) Simplex ProcartaPlex Kit measures CD62E (E-selectin) protein and is designed to be combinable with other Simplex kits, allowing creation of custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (i.e., not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is needed per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on sandwich ELISA principles, using two highly specific antibodies binding to different epitopes of one protein to quantitate multiple protein targets simultaneously on a Luminex instrument.
These multiplex assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and yield analyzed results in approximately four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 μL sample from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex beads used in ProcartaPlex assays are internally dyed with precise proportions of red and infrared fluorophores, creating spectrally unique bead signatures identifiable by Luminex xMAP detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).
Similar to a sandwich ELISA, the assay uses matched antibody pairs to identify the protein of interest. Each bead, labeled with antibodies specific to a single target protein, binds the target and is detected with biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).
Unlike traditional ELISA, the capture antibody in ProcartaPlex assays is conjugated to a bead rather than adsorbed to a microplate well, allowing reagents to remain in solution. Detection is achieved using dual-laser Luminex instruments that identify bead signatures and quantify RPE fluorescence proportional to protein concentration.

ProcartaPlex multiplex assays enable profiling of more target proteins using significantly less sample volume within the same timeframe as a conventional sandwich ELISA. Multiplex panels are available for six species: human, mouse, rat, nonhuman primate, porcine, and canine.
