Target Information

Viral Protein U (Vpu) enhances virion budding by targeting host CD4 and Tetherin/BST2 for proteasomal degradation.
Degradation of CD4 prevents premature interaction between viral Env and host CD4 in the endoplasmic reticulum.
Degradation of Tetherin/BST2 is essential for virion release, as BST2 tethers new viral particles to the host membrane.
Vpu bridges CD4 or BST2 to BTRC, a substrate recognition subunit of the Skp1/Cullin/F-box E3 ubiquitin ligase, inducing ubiquitination and degradation.
Altered E3 ligase specificity by Vpu may promote degradation of host IKBKB, down-regulating NF-κB and inducing apoptosis.
Ion channel activity has been suggested, though its in vivo role remains uncertain.
