Target Information
Isocitrate dehydrogenases catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. These enzymes are divided into two subclasses: NAD(+)-dependent and NADP(+)-dependent.
Among the five known isocitrate dehydrogenases, three are NAD(+)-dependent and located in the mitochondrial matrix, while two are NADP(+)-dependent (one mitochondrial and one cytosolic).
NAD(+)-dependent isocitrate dehydrogenases catalyze the allosterically regulated rate-limiting step of the tricarboxylic acid cycle. Each isozyme is a heterotetramer composed of two alpha, one beta, and one gamma subunit.
The IDH3G gene encodes the gamma subunit of one isozyme of NAD(+)-dependent isocitrate dehydrogenase and is a candidate gene for periventricular heterotopia. Several alternatively spliced transcript variants have been described.
