Target Information
CD31 (platelet endothelial cell adhesion molecule-1, PECAM-1) is an inhibitory coreceptor involved in regulation of T cell and B cell signaling through dual immunoreceptor tyrosine-based inhibitory motifs (ITIM). Upon phosphorylation by associated kinases, these motifs provide docking sites for protein-tyrosine phosphatases.
CD31 is expressed throughout the vascular compartment, predominantly at junctions between adjacent endothelial cells. Its N-terminal Ig-like domain mediates homophilic binding, crucial for cell-cell interactions.
As a multifunctional molecule, CD31 plays diverse roles in:
- Integrin-mediated cell adhesion
- Transendothelial migration
- Angiogenesis
- Apoptosis
- Negative regulation of immunoreceptor signaling
- Autoimmunity and macrophage phagocytosis
- IgE-mediated anaphylaxis and thrombosis
