A recommended positive control tissue is breast carcinoma, though other tissues may also serve as positive controls.
This antibody is intended for laboratory professional use in detecting the corresponding protein in formalin-fixed, paraffin-embedded (FFPE) tissues via manual qualitative immunohistochemistry (IHC). It should be used after the primary diagnosis of tumor has been made by conventional histopathology.
Mammaglobin is a breast-associated glycoprotein belonging to the secretoglobin family, distantly related to uteroglobin and lipophilin. Its expression is highly specific to breast tissue and has been reported as a sensitive marker for occult breast cancer cells in sentinel lymph nodes and peripheral blood. By paraffin IHC, mammaglobin shows approximately 80% sensitivity for breast cancers; when combined with GCDFP-15, overall sensitivity can reach 84%. It is useful in identifying the primary sites of metastatic carcinomas.
For optimal staining, pretreat deparaffinized tissue with heat-induced epitope retrieval or enzymatic retrieval. Standard IHC staining techniques involve sequential application of the primary antibody, secondary antibody, enzyme complex, and chromogenic substrate, followed by microscopic evaluation.
A positive tissue control must be included in every staining procedure. This control should contain both positive and negative staining areas. External positive control materials should be freshly prepared and processed identically to patient specimens. Weakly positive tissues are preferred for quality control to detect subtle reagent degradation or staining variability.
Negative control tissues (internal or external) should also be used according to laboratory policy. Negative control areas should show no specific staining, confirming the absence of nonspecific background signal.
Target Information
Mammaglobin is a gene expressed almost exclusively in normal breast epithelium and human breast cancer. It belongs to the secretoglobin gene family and forms a heterodimer with lipophilin B. Mammaglobin is considered a useful marker in breast cancer research, and its mRNA detection by RT-PCR in peripheral blood correlates with prognostic factors such as lymph node involvement.
