Target Information

MPP8 is a heterochromatin component that specifically recognizes and binds methylated Lys-9 of histone H3 (H3K9me) and promotes recruitment of proteins mediating epigenetic repression. It recruits the HUSH complex to H3K9me3 sites, maintaining transcriptional silencing by promoting SETDB1-mediated H3K9me3 deposition and MORC2 recruitment. MPP8 also recruits DNMT3A to CpG sites, mediating DNA methylation and repressing CDH1 expression. It collaborates with MORC2 and SETDB1 in silencing L1 retrotransposons and unintegrated retroviral DNA, contributing to epigenetic regulation and transcriptional repression.
