Target Information
Calcium-permeable channels, such as TRPV6, participate in neurotransmission, muscle contraction, and exocytosis by providing calcium as an intracellular second messenger. Depending on the tissue, transcellular calcium transport may be regulated by vitamin D, parathyroid hormone (PTH), or calcitonin (CALCA).
TRPV6 acts as a calcium-selective cation channel involved in Ca²⁺ uptake in various tissues, including intestinal reabsorption. The channel is activated by low internal calcium levels and exhibits inward rectification.
Inactivation mechanisms include both rapid Ca²⁺-dependent and slower Ca²⁺-calmodulin-dependent processes, potentially regulated by phosphorylation. In vitro, Mg²⁺ slowly inhibits the channel in a voltage-independent manner.
Heteromeric assembly with TRPV5 modifies channel properties, forming TRPV5-TRPV6 heteromultimers with voltage-dependent gating. TRPV6 interacts with TRPV5, S100A10, ANAX2-S100A10 heterotetramer, BSPRY, and calmodulin. Interaction with S100A10 is required for plasma membrane trafficking.
