Specificity Comments:
Signaling from ligand-activated membrane receptor serine/threonine kinases to nuclear targets is mediated by proteins known as DPC4. Upon ligand binding, TGF-β family receptors phosphorylate SMAD proteins (SMAD1 and SMAD2), which then translocate to the nucleus to activate transcription. This process requires association with SMAD4 (DPC4), a tumor suppressor often inactivated in over half of pancreatic carcinomas. SMAD4 is absent in approximately 80% of pancreatic adenocarcinomas but rarely in other cancers. SMAD4 serves as an important marker for confirming pancreatic adenocarcinoma diagnosis, and its expression correlates with improved patient survival.
Target Information
Smad4 is encoded by the Smad4 gene located on chromosome 18 in humans. It is also known as Smad family member 4, Sma- and Mad-related protein 4, or Mothers against decapentaplegic homolog 4.
Smad4 contains:
- N-terminal MH1 domain: involved in DNA binding
- C-terminal MH2 domain: involved in protein interactions
Binding of TGF-β or BMP ligands to their receptors leads to phosphorylation of R-Smads (Smad1, 2, 3, 5, 8), which then form heterotrimers with Smad4 and translocate to the nucleus.
Smad4 functions as a tumor suppressor by regulating transcription of genes such as Cyclin D1 (downregulation) and collagen (upregulation), thereby inhibiting cell proliferation.
Dephosphorylation regulates the nuclear export and nucleocytoplasmic dynamics of Smads.
