Target Information

This gene generates several transcript variants that differ in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode structurally related isoforms functioning as inhibitors of CDK4 kinase.
Another transcript includes an alternate first exon located 20 Kb upstream, containing an alternate open reading frame (ARF) that produces a structurally unrelated protein. This ARF product stabilizes the tumor suppressor protein p53 by interacting with and sequestering the E3 ubiquitin-protein ligase MDM2, which mediates p53 degradation.
Despite structural and functional differences, both CDK inhibitor isoforms and the ARF product share a common role in G1 cell cycle control through CDK4 and p53 regulation.
Mutations or deletions in this gene are frequently found in various tumors, identifying it as an important tumor suppressor gene.
