Members of the RAS subfamily of GTPases function as molecular switches cycling between inactive GDP- and active GTP-bound states. Guanine nucleotide exchange factors (GEFs), such as Repac (RAPGEF5), activate RAS by promoting GTP binding. RAPGEF5 is a 580 amino acid protein expressed mainly in brain and testis, with weaker expression in heart, placenta, lung, pancreas, and small intestine. It has two isoforms and is conserved across cow, mouse, and rat. The human gene is located on chromosome 7p15.3 and is associated with positive response to erlotinib therapy in non-small-cell lung cancer.
