Target Information
The E3 ubiquitin-protein ligase NEDD4 is a Ubl conjugation pathway protein containing one HECT (E6AP-type E3 ubiquitin-protein ligase) domain and four WW domains. It participates in the degradation of VEGFR-2/KDFR independently of its ubiquitin-ligase activity and acts as a potential proto-oncogene by negatively regulating PTEN via ubiquitination. NEDD4 is also involved in viral budding and regulates epithelial Na⁺ channels, influencing arterial hypertension. It localizes to the sarcolemmal region of muscle fibers, where increased expression is associated with muscle unloading, denervation, and downregulation of Notch1 expression.
