Target Information
Cortactin (Cttn)은 Src의 기질로 처음 확인된 보편적인 액틴 결합 단백질입니다. 이 단백질은 액틴 세포골격과 세포 형태의 조직화에 기여하며, lamellipodia 형성과 세포 이동에도 관여합니다. Cortactin은 세포 주변의 액틴이 풍부한 구조에 축적되어 액틴 네트워크 형성을 촉진하는 역할을 합니다.
Cortactin은 N-말단 산성(NTA) 도메인, 반복 도메인, C-말단의 프롤린-풍부 영역 및 SH3 도메인 등 네 개의 주요 도메인을 가지고 있습니다. NTA는 Arp2/3 및 WASP 복합체와 결합하여 F-actin 가지 형성에 관여합니다. 또한 Cortactin은 세포 이동성과 침습성 촉진에 관여하며, 암세포의 ECM 분해와 관련된 actin-풍부 소세포 돌기(invadopodia)의 형성에도 중요한 역할을 합니다.
Cortactin은 Src 계열 키나아제에 의해 Y421, Y466, Y482 잔기에서 인산화되며, Erk 계열 키나아제에 의해 S405 및 S418 잔기에서 인산화됩니다.
