Target Information
In the intact cell, DNA closely associates with histones and other nuclear proteins to form chromatin. Chromatin remodeling is a critical component of transcriptional regulation, and histone acetylation plays a major role in this process. Acetylation of lysine residues in histones leads to conformational changes that enhance DNA accessibility to transcription factors. Mammalian histone acetylases include GCN5, PCAF, p300/CBP, HAT1, and TAF II p250. Conversely, histone deacetylation is associated with transcriptional silencing. The histone deacetylases (HDAC) family includes HDAC1–9. HDAC9 and HDAC9a are alternatively spliced isoforms, both retaining catalytic activity and repressing MEF2-mediated transcription. They are expressed in brain, skeletal muscle, kidney, placenta, and pancreas.
