Description:
The 104D2 monoclonal antibody reacts with human CD117 (c-Kit), also known as Steel factor receptor and stem cell factor receptor. This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cell subsets and mast cells. Interaction with Steel factor promotes proliferation and differentiation of hematopoietic progenitor cells and mast cell maturation. CD117 is also expressed by melanocytes and plays a role in signaling and activation.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test. A test is defined as the amount of antibody staining a cell sample in 100 µL final volume, with 10⁵–10⁸ cells/test.

Excitation / Emission:
488–561 nm / 578 nm; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

KIT (c-KIT) is a proto-oncogene encoding a type III transmembrane receptor for MGF (mast cell growth factor, also known as stem cell factor). It regulates growth and activation of hematopoietic and non-hematopoietic cells. Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism. Deregulation of KIT (e.g., mutation D816V) is a therapeutic target in neoplastic mast cell growth.

c-Kit is expressed in hematopoietic stem cells, germ cells, mast cells, and gastrointestinal tract Cajal cells. Upon SCF binding, c-Kit dimerizes, activating autophosphorylation of tyrosine residues (e.g., Tyr703), enabling Grb2 docking and Ras/ERK pathway activation. SCF/c-Kit signaling also activates PI3K, PLC-γ, and JAK/STAT pathways, essential for hematopoiesis, stem cell maintenance, and gametogenesis.
