Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 upregulates stress response genes such as p21waf1, leading to G1 or G2/M arrest.

The p53 structure includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 has been associated with tumor formation. p53 functions as a key anticancer agent by blocking proliferation of damaged cells. It also plays roles in aging, oxidative stress, and cognitive processes such as long-term potentiation (LTP).

Mutations in the TP53 gene are common in many cancers and result in loss of tumor suppressor activity. Germline mutations can also occur in cancer-prone families, such as those with Li-Fraumeni syndrome.
