Product Specific Information

Description:
The W6/32 monoclonal antibody reacts with human MHC class I antigens (HLA-A, B, C). These antigens, associated with β2-microglobulin, are expressed on all nucleated cells and play a central role in cell-mediated immune responses and tumor surveillance. The W6/32 clone recognizes a non-polymorphic epitope shared among HLA-A, B, and C loci and immunoprecipitates both 43 kDa and 11–12 kDa chains. Cross-reactivity is also observed in baboon, rhesus, and cynomolgus monkey.

Suitable for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (1.0 µg) per test in a final volume of 100 µL. Cell number may vary (10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass. Ensure instrument compatibility.

Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light during storage and staining.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-Step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss. Clone-specific performance should be verified empirically.

405 nm / 702 nm (Violet Laser)

Target Information

HLA-class I major histocompatibility (MHC) antigens are intrinsic membrane glycoproteins expressed on nucleated cells, noncovalently associated with β2-microglobulin. They present foreign peptides for immune recognition by cytotoxic T cells, playing key roles in antiviral and antitumor defense. HLA-class I molecules include HLA-A, HLA-B, and HLA-C.

HLA and MHC antibodies are essential in Immunopeptidomics, aiding in neoantigen identification and characterization via LC-MS/MS.
