PEX5 binds to the C-terminal PTS1-type tripeptide peroxisomal targeting signal (SKL-type) and is essential for peroxisomal protein import.
Peroxins (PEXs) are critical for the assembly of functional peroxisomes.
Defects in this gene are associated with peroxisomal biogenesis disorders (PBDs), including:

• Neonatal adrenoleukodystrophy (NALD)
• Zellweger syndrome (ZWS)
• Infantile Refsum disease (IRD)

Alternatively spliced transcript variants encoding different isoforms have been identified.
