Target Information

Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. Through this mechanism, ER regulates gene transcription via coactivator or corepressor recruitment.

There are two forms of estrogen receptors, alpha (ESR1) and beta (ESR2), encoded by separate genes. Alternative RNA splicing of ESR1 produces at least four isoforms (66 kDa, 53 kDa, 47 kDa, and 35 kDa). Estrogen receptors are widely expressed and play critical roles in sexual development, reproductive function, bone metabolism, and are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
