Target Information
CNOT1는 CCR4-NOT 복합체의 스캐폴딩 구성 요소로, 세포 내 주요 mRNA deadenylase 중 하나입니다. 이는 mRNA 분해, miRNA 매개 억제, 번역 개시 시의 억제, 일반적인 전사 조절 등 다양한 세포 과정에 관여합니다.
CNOT1의 스캐폴딩 기능은 촉매 복합체 모듈 및 다양한 RNA 결합 단백질과의 상호작용을 통해 특정 mRNA 3’UTR로 복합체를 모집하는 역할을 합니다. 또한 ARE(AU-rich element)를 포함하는 mRNA의 분해에 관여하며, TNRC6A/B/C와의 결합을 통해 CCR4-NOT 복합체를 miRNA 표적 및 RISC 복합체로 모집합니다.
CNOT1은 전사 억제자로 작용하며, 핵 수용체의 리간드 의존적 전사 활성화를 억제하고, 배아줄기세포(ES cell) 정체성 유지에도 관여합니다.
