Target Information
The proteasome is a multicatalytic proteinase complex with a highly ordered ring-shaped 20S core structure. The core consists of four rings of 28 non-identical subunits: two rings of seven alpha subunits and two rings of seven beta subunits. Proteasomes are widely distributed in eukaryotic cells and are responsible for peptide cleavage in an ATP/ubiquitin-dependent, non-lysosomal pathway.
A modified form, the immunoproteasome, processes class I MHC peptides. The PSMB7 gene encodes a member of the proteasome B-type (T1B) family, a 20S core beta subunit. Its expression is downregulated by gamma interferon, and proteolytic processing is required for maturation. This subunit is absent in the immunoproteasome, replaced by catalytic subunit 2i (proteasome beta 10 subunit).
