Target Information

IL-1 alpha (Interleukin-1 alpha) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It is encoded by the IL1A gene and signals through two receptors, IL-1RI and IL-1RII, both shared with IL-1 beta. The IL1A gene is located on chromosome 2q13.

IL-1 alpha plays a key role in innate host defense by inducing other proinflammatory cytokines and initiating acute-phase responses. Its activity can be moderated by IL-1 Receptor Antagonist (IL-1RA), which blocks receptor binding through competitive inhibition.

This cytokine is involved in immune responses, inflammation, and hematopoiesis. It is produced as a proprotein by monocytes and macrophages, processed upon cell injury, and can induce apoptosis. IL1A and related interleukin genes form a cytokine cluster on chromosome 2. IL-1 alpha regulates NF-κB and MAPK pathways, stimulates IL6 and COX-2 (PTGS2) expression, and enhances nitric oxide (NO) production.

Elevated IL-1 alpha levels are associated with chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis.
