Target Information
Guanylate kinase (GK) catalyzes the ATP-dependent phosphorylation of GMP into GDP, essential for recycling GMP and indirectly cGMP. It is a conserved monomeric protein (~200 amino acids) found in prokaryotes, lower eukaryotes, and vertebrates. GK shares structural similarity with Vaccinia virus protein A57R (SalG2R). Proteins containing DHR, SH3, and GK-like domains are termed MAGUKs (membrane-associated guanylate kinase homologs), including Drosophila dlg1, mammalian Zo-1, SAP90/PSD-95, CHAPSYN-110/PSD-93, SAP97/DLG1, SAP102, erythrocyte p55, C. elegans lin-2, rat CASK, and human DLG2, DLG3. GK has an ATP-binding site (P-loop) in the N-terminal region, which is not conserved in GK-like domains of MAGUKs, though key binding residues remain.
