Target Information
G protein-coupled receptor GPR49 (LGR5) is expressed in brain, skeletal muscle, placenta, and spinal cord. ESTs have been isolated from normal human brain (amygdala), embryo, and placenta libraries and from human germ cell, uterus, and brain libraries.
G-protein Coupled Receptors (GPCRs) are one of the largest families of signaling molecules, characterized by seven membrane-spanning helices and existing in both active and inactive forms. Ligand binding induces conformational changes leading to G-protein signaling and downstream effects.
Negative regulation ("turn-off" activity) of GPCRs plays an essential role in cellular physiology and pharmacological research. GPCR-targeted drugs such as Zyprexa (Eli Lilly) and Claritin (Schering-Plough) are major pharmaceuticals in mental health and allergy treatment.
