Target Information

Proposed scaffold protein implicated in diverse physiological processes, partly linked to regulation of ubiquitination of specific cellular proteins. Modulates activity of cullin-RING E3 ubiquitin ligase (CRL) complexes by displacing CAND1, promoting CRL E3 activity and dissociating CAND1 from CUL1 and CUL2. Promotes ubiquitination and proteasomal degradation of NF-kappa-B subunit RELA, leading to down-regulation of NF-kappa-B activity. Regulates membrane expression and ubiquitination of SLC12A2 and modulates Na(+) transport in epithelial cells through ENaC subunit regulation. Promotes localization of SCNN1D to recycling endosomes and enhances CFTR surface expression. Involved in copper ion homeostasis and copper-dependent ATP7A trafficking, functioning within the CCC complex and cooperating with the WASH complex. Binds one copper ion per monomer and phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2). Also regulates HIF1A-mediated transcription by competing with ARNT/Hif-1-beta, resulting in decreased DNA binding and impaired transcriptional activation.
