Product Specific Information

This product is provided in a ready-to-use formulation.
A recommended positive control tissue is tonsil, though other positive controls may also be used.

The primary antibody is intended for professional laboratory use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissue using manual qualitative IHC testing. It should be used after the primary diagnosis of a tumor has been made by conventional histopathology.

FOXP1 (Forkhead box protein P1) is a transcriptional repressor involved in the specification and differentiation of lung epithelium and acts cooperatively with FOXP4 to regulate epithelial cell fate. It is also an essential regulator of B cell development. FOXP1 is useful in the subclassification of DLBCL, with a high cutoff (≥80%) required for high specificity for the ABC subtype.

For optimal results, pretreat deparaffinized tissue with heat-induced or enzymatic epitope retrieval.
IHC staining enables visualization of antigens through sequential application of primary and secondary antibodies, enzyme complexes, and chromogenic substrates. The resulting signal is observed under a light microscope to assist in differential diagnosis.

A positive tissue control must accompany every staining procedure.

  • Control tissues should contain both positive and negative staining areas.
  • External positive control materials should be fresh, properly fixed, and embedded specimens.
  • Weakly positive tissues are preferred for quality control and detecting reagent degradation.

Negative controls (internal or external) should confirm the absence of nonspecific staining. If specific staining appears in negative controls, results from patient specimens must be considered invalid.

Target Information

The FOXP1 gene belongs to the forkhead box (FOX) transcription factor family, which regulates tissue- and cell type-specific gene expression during development and adulthood. FOXP1 contains DNA-binding and protein–protein interaction domains and may function as a tumor suppressor. It is located on chromosome 3p14.1, a region linked to tumor suppressor activity. Alternative splicing produces multiple isoforms.
