cGAS (Cyclic GMP-AMP synthase)
cGAS는 ATP와 GTP로부터 cyclic GMP-AMP (cGAMP)를 형성하는 nucleotidyltransferase로, 선천면역에서 핵심적인 역할을 수행합니다. 이 효소는 GpA 단계에서 2’,5’ 인산다이에스터 결합, ApG 단계에서 3’,5’ 인산다이에스터 결합을 형성하여 c[G(2’,5’)pA(3’,5’)p]를 생성합니다.
cGAS는 세포질 DNA에 직접 결합하여 cGAMP 합성을 유도하고, 이는 TMEM173/STING을 활성화시켜 type-I interferon 생성을 유도합니다. 또한 감염의 선천면역 감지, 세포 스트레스 및 DNA 손상 반응, 세포 노화 조절에도 관여합니다.
