Target Information
Semaphorins constitute the largest family of axon guidance cues, divided into eight classes (3–7 in vertebrates).
- Class 3 Semaphorins: Secreted proteins
- Classes 4–6: Transmembrane proteins
- Class 7: Membrane-associated via GPI linkage
They share a conserved ~400 amino acid sema domain and function as axon repulsion mediators or chemoattractants depending on context.
Semaphorins also play roles in cardiovascular development and immune cell antigen presentation.
Their signaling is mediated by Neuropilin and Plexin receptor families.
