Target Information

The Ras superfamily of small GTPases includes the Ras, Rho, Arf, Rab, and Ran families. Ras-regulated signaling pathways control processes such as actin cytoskeletal integrity, proliferation, differentiation, cell adhesion, apoptosis, and cell migration. These GTPases function as molecular switches that regulate eukaryotic cell function by cycling between a GDP-bound inactive form and a GTP-bound active form. GTP binding induces conformational changes in the effector-binding domain of the G-protein.

Ras proteins are monomeric G-proteins with molecular masses of 20–30 kDa and are attached to the cell membrane by prenylation. Mutations in Ras family proto-oncogenes (H-Ras, N-Ras, K-Ras) are among the most common in human cancers, occurring in 20–30% of tumors. K-Ras mutations are frequent in non-small-cell lung, colorectal, and pancreatic carcinomas; H-Ras mutations in bladder, kidney, and thyroid carcinomas; and N-Ras mutations in melanoma, hepatocellular carcinoma, and hematologic malignancies.
